Background and applications

Multidimensional Liquid Chromatography (MDLC)

This is a multidimensional liquid chromatography (MDLC) system with wide dynamic range. MDLC provides micropreparative protein and peptide prefractionation from complex mixtures as well as nanoscale peptide separation. It enables the analysis of hydrophobic membrane proteins, proteins with extremes in size as well as extremely basic and acidic proteins. The flow paths in the MDLC are made of bio-inert materials, to ensure high recovery of labile peptides (e.g. phosphopeptides) and enable analysis of post-translational modifications. It delivers accurate and reproducible flow rates and gradients, with flow rate ranging from 200 nl/min to 2 ml/min. The MDLC system interfaces with the LTQ mass spectrometer to enable rapid protein identification, and is integrated into and operated from Thermo Electron's Xcalibur™ software.

Instrument specs

Some of the MDLC features include:

  • Unique conductivity flow cell – real time monitoring of on-column gradient profile
  • High pressure gradient mixing – improved resolution, accuracy and reproducibility
  • High throughput and dynamic range – matching the outstanding speed and sensitivity of the LTQ
  • Wide flow rate range and dual functionality– protein and peptide pre-fraction and nanoflow peptide separation
  • Bio-compatible flow path – essential for analyses of low abundant proteins and PTMs
  • Patented low volume nanovalves – for maximum sensitivity and resolution
  • High flow accuracy – extremely reproducible retention times
  • Pre-configured software methods – simplified system operation with the option to customize own methods
  • Autosampler – Peltier cooled, 96-384 microplate samples inject 0.1 ul – 500 µl
